魚類免疫系を活用した一本鎖抗体取得システムの開発 Development of the system for obtaining a single-chain antibody using goldfish immunity 田丸, 浩 Tamaru, Yutaka 一本鎖抗体 キンギョ ゼブラフィッシュ ファージディスプレイ application/pdf 3コピーのhLGR5遺伝子を搭載したタンデムリピート型プラスミドをゼブラフィッシュ受精卵にマイクロインジェクションすることで、hLGR5タンパク質を発現することに成功した（特許出願１件）。 次に、キンギョIg遺伝子についてコイ科およびニジマスのゲノム情報と比較した結果、キンギョゲノムにはIgM遺伝子に加えて、IgZ、IgZ2、IgZ3遺伝子の存在が確認された。さらに、大腸菌曝露前後におけるキンギョIg遺伝子発現量の変動を確認した結果、キンギョ（和金）ではIgZ、IgZ2は鰓でのみ遺伝子発現量が増加し、IgM遺伝子はゼブラフィッシュほどの発現増加は検出されなかった。 A tandem repeat of plasmid carrying three hLGR5 genes was injected into zebrafish fertilized eggs. As a result, the genes encoding IgZ, IgZ2, and IgZ3 in goldfish genome was found in addition to IgM. Furthermore, the gene expression pattern among those Igs was observed before or after exposing Escherichia coli.
