ヒト剖検脳とモデルマウスを用いた脳アミロイド血管症の病態解明 Elucidation of the pathogenesis of cerebral amyloid angiopathy using model mice. 水谷, あかね Mizutani, Akane 脳アミロイド血管症 CAA application/pdf 脳アミロイド血管症(CAA)は加齢に伴い増加し、血管性認知症の発症に関与するほか、アミロイドのクリアランスを低下させアルツハイマー病の進行にも関与する。本研究はCAAモデルマウスを用いてCAAの発症機序を解明すること目的としている。まず、CAAモデルマウスにおいて、脳血管の基底膜の内側にAPP770の発現を確認したが、Aβの沈着は確認されなかった。慢性低灌流負荷を加えたところ出血が増加することを確認したが、ウェスタンブロッティングでは炎症関連分子の発現に有意差は見られなかった。今後、慢性低還流負荷による出血増加の機序の解明や、治療介入により病態改善がみられるか確認していく予定である。 Cerebral amyloid angiopathy (CAA) increases with aging and is involved in the onset of vascular dementia, as well as in the progression of Alzheimer's disease due to decreased amyloid clearance. The purpose of this study is to elucidate the pathology of CAA using CAA model mice. First, in CAA model mice, expression of APP770 was confirmed inside the basement membrane of cerebral vessels, but deposition of Aβ was not confirmed. Chronic hypoperfusion stress increased bleeding, but Western blotting showed no significant difference in the expression of inflammation-related molecules.
