{"created":"2023-06-19T11:41:47.897730+00:00","id":10718,"links":{},"metadata":{"_buckets":{"deposit":"6c76f528-7afc-477e-8ae0-9d580653db4c"},"_deposit":{"created_by":15,"id":"10718","owners":[15],"pid":{"revision_id":0,"type":"depid","value":"10718"},"status":"published"},"_oai":{"id":"oai:mie-u.repo.nii.ac.jp:00010718","sets":["420:724:1692754926352"]},"author_link":["26789"],"item_8_biblio_info_6":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2007-03-01","bibliographicIssueDateType":"Issued"}}]},"item_8_description_14":{"attribute_name":"フォーマット","attribute_value_mlt":[{"subitem_description":"application/pdf","subitem_description_type":"Other"}]},"item_8_description_4":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"植物の自家不和合性は、雌蕊と花粉との間の自他認識反応により、自己花粉の受精を抑制し他家受粉を促進する性質である。サツマイモを含むIpomoea属植物は胞子体型自家不和合性を有しており、単一遺伝子座(S遺伝子座)の複対立遺伝子によってその自他認識反応が制御されている。本研究は、S遺伝子座にコードされている遺伝子を同定し、自家不和合性の分子遺伝学的機構を明らかにすることを目的としている。本年度の研究成果の要点は、次の通りである。(1)S_1とS_<10>ハプロタイプのS遺伝子座領域について、ゲノム塩基配列を解読し比較した結果、塩基配列の多型性が高い約30〜50kbpの変異領域が明らかになり、S遺伝子座の物理的範囲がこのように狭い範囲内に限定された。(2)この変異領域には、3種類の柱頭特異的発現遺伝子(SE-1,-2,-A)と3種類の葯(花粉)特異的発現遺伝子(AB-1,-2,-3)が座乗していることが明らかになった.(3)これらの遺伝子がコードする蛋白質のアミノ酸配列をSハプロタイプ間で比較した結果、AB-1とAB-3では95%以上の相同性が認められたが、これら以外の遺伝子では高い多型性が認められたことから、AB-2と3種類の柱頭特異的遺伝子は有力なS遺伝子候補であることが確認された。(4)データベース検索から、AB-2遺伝子は花粉外被蛋白質(pollen coat protein)をコードしていると推定されたが、柱頭特異的遺伝子では有意な相同性遺伝子は見出されなかった。(5)葯特異的遺伝子(AB-2)および柱頭特異的遺伝子(SE-2,SE-A)について,RNAiコンストラクトを設計し形質転換体の作出を行った。いずれのコンストラクトでも1〜2個体の形質転換体が得られているが、今後さらに多数の形質転換体を作出し自家不和合性形質の変化を調査する必要がある。これらの実験から、Ipomoea属植物の自家不和合性に関与する遺伝子が間もなく同定されると期待される。","subitem_description_type":"Abstract"}]},"item_8_description_5":{"attribute_name":"内容記述","attribute_value_mlt":[{"subitem_description":"津","subitem_description_type":"Other"},{"subitem_description":"57","subitem_description_type":"Other"}]},"item_8_description_62":{"attribute_name":"目次","attribute_value_mlt":[{"subitem_description":"Genetic
