| 内容記述 | ニワトリ砂嚢から調製した平滑筋γ-アクチンのATPやCaイオンに対する親和性を測定した。EDTAをアクチン溶液に添加してCaイオンをキレートすると、平滑筋γ-アクチンは骨格筋α-アクチンに比して速やかに変性した。みかけの変性速度の運動学的解析から、平滑筋γ-アクチンおよび骨格筋α-アクチンのATPに対する結合定数は、それぞれ1.6×10^3 M^-1、4.7×10^4 M^-1であることが明らかになった。蛍光性カルシウム指示薬であるQuin2とアクチンの競合結合実験からは、Caイオンに対する親和性も平滑筋γ-アクチンの方が約8.5倍低いことが示された。したがって、平滑筋γ-アクチンの不安定性は、構造安定化因子であるヌクレオチドや2価イオンとの親和性の低さに起因することが明らかになった。 | |||||||||||||

| 内容記述 | We investigated ATP and Ca2+ binding to smooth muscle γ-actin prepared from chicken gizzard. When bound Ca2+ was removed with EDTA, smooth muscle γ-actin denatured more rapidly than skeletal muscle α-actin. Kinetic studies of the denaturation process showed that the binding constants of ATP to smooth muscle γ-actin and skeletal muscle α-actin were 1.6×103 M-1 and 4.7×104 M-1, respectively. Competitive binding of Ca2+ between actin and Quin2, a fluorescent Ca2+ indicator, showed that affinity of Ca2+ for smooth muscle γ-actin was also lower than that of skeletal muscle α-actin by a factor of 8.5. These results indicated that smooth muscle γ-actin was relatively unstable due to the low affinities of these stabilizing ligands. | |||||||||||||
