Samsun required a srtong light and suitable plant growth regulators and carbon sources for its growth. Little growth of the callus tissue was observed on the medium containing sugar in the dark or on the sugar-free medium under the strong light condition. The sugar-and light-dependent growth of the callus tissue was completely inhibited by 10⁻⁶ M of DCMU. Callus cells possess many chloroplasts with well-developed grana, but the activity of RuDP carboxylase of the callus tissue was far less than the enzyme activity of intact plant leaves. This callus tissue grew in proportion to some concentrations below 3% of sugar in the medium.
