Total RNA was extracted from CMV-infected leaves of Nicotiana glutinosa by the simplified SDS/phenol method. The RNA was initially reverse-transcribed into cDNA, then the CMV coat protein‐coding sequence was amplified by the PCR method. The detection limit of CMV by the RT‐PCR method ranged from 10 to 100 ng of CMV particles per 10 mg of fresh leaf weight.
