Delta-1、GM-CSF、TGF-β1の存在下でLangerhans細胞に分化することを見出した。これらの細胞はCD1a、E- Cadherin、Langerin、CCR6を発現し、Birbeck顆粒を有していた。また、貪食能とMIP-3αに対する走化性を示した。TNF- αとCD40リガンドで分化させると、MIP-3βに対する走化性を獲得し、CD4陽性T細胞とCD8陽性T細胞を刺激した。GM-CSFとIL-3はシグナル伝達に関与する受容体subunitを共有するが、IL-3は皮膚環境において構成的には産生されていない。そこで、末梢血単球の Langerhans細胞への分化におけるGM-CSFの皮膚環境での位置づけをより明確にするために、GM-CSFとIL-3を比較検討した。GM- CSFをIL-3に替えて培養すると、末梢血単球はLangerhans細胞へ分化しなかった。末梢血単球のLangerhans細胞へ分化におけるGM -CSFの不可欠性を示している。予備的実験において、遺伝子の発現プロファイルをmicroarrayで解析したところ、GM-CSF+TGF-β1+
