{"created":"2023-06-19T11:39:47.790234+00:00","id":7976,"links":{},"metadata":{"_buckets":{"deposit":"17255d0b-0df6-418a-8cc7-7f8e9db8d7b6"},"_deposit":{"created_by":15,"id":"7976","owners":[15],"pid":{"revision_id":0,"type":"depid","value":"7976"},"status":"published"},"_oai":{"id":"oai:mie-u.repo.nii.ac.jp:00007976","sets":["334:627:1692670222110"]},"author_link":["19342"],"item_8_biblio_info_6":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2008-05-01","bibliographicIssueDateType":"Issued"}}]},"item_8_description_14":{"attribute_name":"フォーマット","attribute_value_mlt":[{"subitem_description":"application/pdf","subitem_description_type":"Other"}]},"item_8_description_4":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"肝線維化に深く関与する肝星細胞が造血幹細胞に由来するとの仮説を立て骨髄移植モデルを用いて検証した。EGFPマウスの骨髄からlineage陰性c-kit陽性sca-1陽性CD34陰性細胞を1細胞ずつソートし、SCF+IL-11存在下で7日間培養した。その時点で20細胞以下の細胞を含むwellから全細胞を回収し、Ly-5.1マウスのsca-1陰性骨髄単核球と一緒に9.5Gy照射済Ly-5.1マウスに移植した。移植2ヶ月後の末梢血の解析から、移植した60匹中7匹(12%)でlong-term, multilineage engraftmentが確認された。これらのマウスに週2回12週以上にわたり四塩化炭素を腹腔内注入した。マウスから摘出した肝臓は表面凹凸不整でAzan染色標本にて線維化が示された。肝臓内にはEGFP陽性細胞が数多く存在し、その細胞の表現型を免疫組織染色にて検討した。50-60%のEGFP陽性細胞はCD45陰性で、細胞内に脂肪滴を有していた(oil-red-O陽性)。このEGFP陽性oil-red-O陽性細胞はvimentinとGFAPが陽性であった。一部のEGFP陽性細胞ではα-smooth muscle actinが陽性であった。肝臓を酵素処理後3日間10%FCS加DMEM下に培養して得たEGFP陽性細胞はoil-red-O陽性で、collagen-IとADAMTS13を産生し、肝星細胞の特徴を有していた。この結果が、ドナー由来造血幹細胞とレシピエントの肝星細胞との融合によるものかどうかをY-chromosome
