{"created":"2023-06-19T11:39:54.720253+00:00","id":8139,"links":{},"metadata":{"_buckets":{"deposit":"2f9bce90-2275-4d3c-b665-b25b218ce645"},"_deposit":{"created_by":15,"id":"8139","owners":[15],"pid":{"revision_id":0,"type":"depid","value":"8139"},"status":"published"},"_oai":{"id":"oai:mie-u.repo.nii.ac.jp:00008139","sets":["631:636:1692684609508"]},"author_link":["20207","20208","20209","20210"],"item_8_biblio_info_6":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2013-05-28","bibliographicIssueDateType":"Issued"}}]},"item_8_description_14":{"attribute_name":"フォーマット","attribute_value_mlt":[{"subitem_description":"application/pdf","subitem_description_type":"Other"}]},"item_8_description_4":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"我々は化学放射線療法に抵抗し遺残した癌細胞の中に癌幹細胞が多く含まれているとの仮説を立て、遺残癌細胞の分子生物学的特徴が直腸癌化学放射線療法後の遠隔転移再発や予後に関連し、術前化学放射線療法によって刺激を受けた癌細胞の一部が上皮間葉移行をきたし転移能を獲得しているのではないかと考えた。化学放射線療法に抵抗性を示したと考えられる遺残癌細胞では、CD133やLgr5(幹細胞マーカー)、GLUT1(糖代謝)やHIF1a(組織低酸素)の発現は再発や予後不良と関連していた。またTSやDPD(5-FU抵抗性マーカー)の発現は再発と関連していた。遺残癌細胞周囲の間質細胞では、CXCR4やCXCL12(癌増殖、浸潤、転移と関連)やFAP-αやSDF-1(cancer-associated fibroblasts関連)の発現は再発や予後と関連していた。Neurotrophic receptor tropomyosin related kinase (TrkB)高発現大腸癌ではE-cadherinの発現が低く予後不良であり、TrkBが大腸癌細胞の上皮間葉移行を誘導した。放射線照射が大腸癌細胞の上皮間葉移行を誘導し、移動能、浸潤能を増強した。放射線照射後の生存(遺残)細胞ではCD133の発現が上昇し、放射線照射が大腸癌細胞の上皮間葉移行を誘導し、照射後遺残細胞にCD133発現陽性細胞が多く存在していることが示唆された。今後も、大腸癌肝転移巣における癌幹細胞の振る舞いや癌幹細胞の上皮間葉移行の関与を形態学的および分子生物学的に検討する必要がある。 ","subitem_description_type":"Abstract"},{"subitem_description":"We hypothesized that residual cancer cells after chemoradiotherapy (CRT) for rectal cancer have the treatment (CRT)-resistant property and contain putative CSCs, compared with pre-CRT cancer specimens. Furthermore, residual cancer cells which survived CRT may acquire metastatic potential induced by EMT. The expression of CD133, leucine-rich repeat-containing G protein-coupled receptor 5 (LGR5), glucose transporter 1 (GLUT1), and hypoxia inducible factor-1α (HIF-1α) on residual cancer cells after CRT was associated with cancer recurrence and patient prognosis. The expression of thymidylate synthase (TS) and dihydropyrimidine dehydrogenase (DPD) on residual cancer cells after CRT was also associated with cancer recurrence. The expression of CXCR4 and its ligand CXCL12, fibroblast activation protein-α (FAP-α), and stromal cell-derived factor-1 (SDF-1) on the cancer stroma after CRT was also associated with cancer recurrence and patient prognosis. In vitro study, radiation induced EMT in colorectal cancer cells, resulting in the acquisition of invasive and metastatic properties. Neurotrophic receptor tropomyosin related kinase (TrkB) also induced EMT in colorectal cancer cells. TrkB was inversely correlated with E-cadherin, and associated with poor prognosis of colorectal cancer patients.
