| 内容記述 | 毛顎動物であるヤムシの精子は、精巣より遊離した精原細胞が尾部体腔内を循環しながら増殖・成長することにより形成される。この精子形成過程を明らかにするために、体腔から摘出した様々なステージの細胞を容易に観察できる試料作成法について検討した。体腔液をスライドグラスにとり、空気乾燥後にメタノール固定した標本を酢酸オルセインまたはヘキスト33342で核染色したところ、良好な観察像が得られた。また、この標本では、免疫組織化学染色も可能であったことから、ヤムシ精子形成過程を研究するための標本作成法として有用であることが示された。 | |||||||||||||

| 内容記述 | Chaetognaths are hermaphrodites and the male reproductive organs are located in the tail coelom. Spermatogonia released from the testes develop while circulating in the tail coelom. To understand the process of spermatogenesis,we tried to make preparations for observation of various stages of spermatogenic cells. The tail coelomic fluid put on a glass slide was completely dried and fixed with methanol, giving good profiles by staining with acetic orcein or Hoechist33342 dye. This preparation was also available to immunocytochemical staining, indicating that it is useful for the study of spermatogenesis in chaetognath. | |||||||||||||
