All creatures, including procaryotes, harbor genetic information in deoxyribonucleic acid (DNA). Some viruses, however, store their genes in the form of ribonucleic acid (RNA) instead of DNA. To elucidate the functions of RNA viruses, determination of the nucleotide sequences making up their genetic material is necessary. However, direct sequencing of RNA is at present impossible. To address this problem, synthesis of complementary DNA (cDNA) using reverse transcriptase is generally applied. The problem is that the efficiency of this reverse reaction is extremely low. We have therefore employed the polymerase chain reaction to amplify cDNA after reverse transcription, and could thereby successfully determine the complete nucleotide sequence of a polyhedrin gene of Bombyx mori cytoplasmic polyhedrosis virus, which harbors its genome as double-stranded RNA. |
